Just to prevent sucking air at the joint, is why I would figure using sealer. TEST FITTING is an excellent idea! You should be able to screw the pick-up in by hand, to within a quarter turn. That last quarter should be with a pipe wrench, and only once or twice. Too many test fittings will loosen the seal between the two threads. You need to verify the pick-up screen is parallel with the block rails, and the pan will fit on too! I would try to fit the pick-up screen and pan without a gasket, and JUST touch each other. Installing a gasket or windage tray at final assembly will give you the clearance you need. You probably knew that already, but just in case, don't forget.
