I just found this Tech video on the "59" torqueflite

It talks about how its all new. It did bring up something I have not heard about though.

It mentions a spring in the valve body that is not present on a car equipped with a 2x4.

Anyone aware of this and what it does?

I am going to be rebuilding the transmission soon in my 60 plymouth and I am planning a 2x4 (maybe not right away but not to far into its road going future) and
wonder if I should remove this spring or not.

Whats the thoughts on this

They're talking about the accumulator spring, one of the most misunderstood parts in the TF.

The accumulator absorbs the shock created by rapid fluid movement both on the 1-2 up-shift and on the initial shift to Drive. There is a belief that the presence of the spring "softens" the 1-2 up-shift but it doesn't and the belief is that removing the spring "firms" the 1-2 up-shift...again, it doesn't. The absence of the spring puts extra strain on the rear clutch so I have no idea why the factory left it out on performance installations all the way up into the seventies.
